Description
Seroquel [Quetiapine] is an oral medication found in a form of tablets of different strengths 25mg 50mg 100mg 200mg 300mg and 400mg. It has been introduced in so many strengths to increase dosing flexibility. Seroquel [Quetiapine] is composed of an active ingredient called Quetiapine and other inactive ingredients. Seroquel [Quetiapine] is an antipsychotic drug used for the treatment of schizophrenia depression and mania associated with bipolar disorder. Off-label uses of Seroquel are obsessive-compulsive disorder autism alcoholism neuropsychiatric disorder post-traumatic stress disorder and restless legs syndrome. Physicians sometimes use Seroquel [Quetiapine] as a sedative to patients having sleep disorder and anxiety disorder. It is thought that Seroquel has a sedative effect because quetiapine present in it has an antagonistic effect on the histamine H1 receptor. It is thought that Seroquel [Quetiapine] has the beneficial effect because it blocks the activity of dopamine receptors specifically D1 and D2 dopamine receptor alpha-1 and alpha-2 adrenergic receptor and serotonin receptors. By blocking these receptors Seroquel inhibits certain chemicals known as neurotransmitters used to communicate with neurons and nerve cells.
How to use
The recommended initial dose of Seroquel [Quetiapine] for the treatment of schizophrenia is 25mg daily which will increase by 25-50mg daily to 300mg or 400mg daily on day 4. Seroquel generally produce desired effect for the said treatment on the dosage ranging between 150mg to 750mg/day.
The recommended initial dose of Seroquel for the treatment of depression associated with bipolar disorder is 50mg once daily at bedtime which will increase to 100mg to 200mg and to 300mg on the day 4. It can be further increased to 400mg on day 5 and to 600mg by day 8 for the desired effect. But no benefits are recorded in consuming 600mg than 400mg.
The recommended initial dose of Seroquel for the treatment of mania associated with bipolar disorder is 100mg daily in bid doses. It can be increased to 400mg by day 4 with an increment of 100mg/day. For the desired effect the dose can be increased to 600mg to 800mg on day 6. But the incremental dose should not exceed 200mg/day. Majority of patients respond between 400mg and 800mg.


The initial dose for bipolar mania is 50 mg twice daily 100 mg/d. The dose can be increased by 100 mg/d to a daily dose of 400 mg/d. Most patients respond to 400-800 mg/d. Doses greater than 800 mg/d have not been studied.

The initial dose for schizophrenia is 25 mg twice daily 50 mg/d. The dose can be increased by 25-50 mg two or three times daily. The target dose is 300-400 mg/d in two or three doses. Patients respond to 150-750 mg/d and doses greater than 800 mg/d have not been evaluated.
Side Effects
Seroquel [Quetiapine] has side effects which are common serious and life-threatening too. The most common side effect of Seroquel is sedation. It is one of the most sedating of all antipsychotic drugs. Because of its sedating effect Seroquel used for bipolar disorder treatment is recommended to be administered at bedtime which disappears after some time or with the change of dosage.
Other common side effects of Seroquel are weight gain constipation headache dry mouth and a rapid heart beating during the time of inactive or sleeping stage which is known as Tachycardia.
Even less common side effects are stuffy nose stomach disorder abnormal liver disorder dizziness restlessness and paranoia fear and anxiety to the level of irrationality and delusion.
Prolonged use of Seroquel and some other neuroleptic drugs may develop a risk of incurable Tardive dyskinesia involuntary repetitive and purposeless movements.
Use of Seroquel can result in life-threatening neurological disorder known as Neuroleptic malignant syndrome though it’s rare.

Drug Interaction
Seroquel [Quetiapine] acts as sedative should not be co-administered with other sedative drugs or drugs this with sedative effects which will result in increase of sedating effect.
Quetiapine present in Seroquel can cause hypotension or low blood pressure and therefore increase the blood pressure lowering effects of antihypertensive drugs.
Patients taking phenytoin may require higher doses of Seroquel because phenytoin absorbs quetiapine from intestine and result in very low effect of quetiapine.
Enzyme in the liver called cytochrome P450 3A eliminates quetiapine from the body. So drugs interfering with this enzyme when taken with Seroquel may cause increased and toxic level of quetiapine.
Uses
Seroquel [Quetiapine] drug is officially used for the treatment of schizophrenia depression and mania associated with bipolar disorder.
Quetiapine present in Seroquel is used to minimize the symptoms of opioid withdrawal. It has produced desired effect on patients suffering sleep disorder anxiety disorder major depressive disorder obsessive-compulsive disorder post-traumatic disorder autism restless legs syndrome and alcoholism.
Storage
Seroquel [Quetiapine] should be stored at a room temperature between 15 o to 30 oC 59 o to 86 oF away from light heat and moisture.
